Natural antibodies have been shown to mediate the in vivo response to ischemia-reperfusion (IR) injury, including protection from intestinal IR injury. The present studies establish that the annexin IV-recognizing IgM natural antibody, mAb 150, is required for IR-induced protection in mice. mAb 150 recognizes a neo-epitope of annexin IV that is exposed in IR-induced damage to the intestinal epithelial cells, as evidenced by its presence in high levels in the ischemic jejunum and colon. These results indicate that annexin IV is a major IR-related neo-epitope that is recognized by pathogenic natural Abs and is an essential component of the natural Ab repertoire. Further exploration of the mechanisms that drive natural Ab self-reactivity to annexin IV, determination of how and where annexin IV epitopes are displayed on cells, and the understanding of how this part of the natural antibody repertoire is positively selected by CR2 are all areas for future investigation.
